Cloning of FliL and FliM Genes of the Flagellar Operon of Photosynthetic Purple Sulfur Bacterium Allochromatium vinosum

Objective: The goal of this study was to clone and sequence some of the flagellar genes of photosynthetic purple sulphur bacterium Allochromatium vinosum and to perform phylogenetic analysis.

Materials and Methods: Molecular biology techniques such as DNA isolation, restriction digestion, ligation, polymerase chain reaction (PCR), bacterial transformation and DNA sequencing as well as bioinformatic tools such as Clustal W Aligment, NCBI Blast, MEGA software were used throughout the study.

Results: In this study we reported the cloning and sequencing of FliL and FliM genes from the flagellar gene cluster of A. vinosum and discussed the results via parsimony analysis. According to the parsimony analysis of FliL and FliM genes, A. vinosum falls into gamaproteobacteria which also includes nitrogen fixing bacterium A. vinelandii.

Conclusion: For the functional characterization of FliL and FliM genes, mutation analysis of the related sequences are needed.
