Comparison of Results of Urine Cultures and a Flow Cytometric Device That Analyses Urinary Leukocytes/ Bacteria

Objective: It was aimed to make a fast screening algorithm by comparing the results obtained by the flow cytometry method with the urine culture results so as to to eliminate cultivation in samples containing a number of leukocytes and bacteria below a threshold value. In the second step of the study; it was evaluated to what extent unnecessary antibiotic usage could be avoided by finalizing the results of negative-contaminated urine specimens in a short-term.

Material and Methods: The urine samples of 995 patients from all clinics submitted to our laboratory for culture were also tested in a flow cytometry device. The results obtained from the flow cytometry device were compared with the culture results. In the second step, the use of antibiotics in 208 patients referred from the urology clinic were recorded, in order to evaluate the empirical use of antibiotics, which were initiated without the culture results were obtained.

Results: The results we obtained with the flow cytometry method showed that there was no need to carry out urine culture in 31% of our samples that were sent from all the clinics. As to the samples obtained from the urology clinic, it was detected that there was no need to carry out urine culture in 29.3%; and 23% of the patients whose urine cultures were detected as “no growth” on flow cytometry predicted had been started on unnecessary antibiotic therapy.

Conclusion: When this technique is used as a rapid screening test, it will reduce the workload and cost in laboratories in which urine specimens are frequently sent for analysis; also contribute to a certain extent in reducing unnecessary antibiotic use by providing negative results within minutes.
