In Vytro Effects Of Ampycyllyn, Cyprofloxacyn And Ofloxacyn On Salmonella Typhy Wythyn Human Monocyte- Deryved Macrophages

Enteric fever is still a serious health problem in most developing countries. Like other infections caused by intracellular pathogens, growing ability of Salmonella species in phagocytes is an important pathogenicity criteria. By entering host cell, they grow in phagolisosomes and are protected from effects of antibacterial agents, thus the effectiveness of antibiotics can not be good in-vitro as in-vivo.

In this study, we examined the killing effects of ampicillin, ciprofloxacin and ofloxacin to two Salmonella typhi strains (clinical and a standard), within human monocyte-derived macrophages. Human monocyte derived macrophages were infected by two S. typhi strains at a cell ratio of 10 bacteria per one macrophage (10:1). Antibiotics were added at MIC and 10 X MIC. Specimens were examined by both culture and LIVE/DEAD BacLight staining system which allow us to see dead and live bacteria directly. At MIC the least potent intracellular agent was ampicillin and at 10 X MIC all three had good activity. When specimens were estimated by using BacLight staining system, made possible us to see directly live bacteria, which could not be detected by culture method. All three antibiotics had good effects on intracellular S. typhi at 10 X MIC, whereas ampicillin had not at MIC.
