THE EFFECTS OF ANTIPSEUDOMONAL DRUGS AND THEIR COMIBINATIONS ON THE POLYMORPHPNUCLEAR LEUCOCYTE FUNCTIONS |

In our study the eff ects of antipseudomonal drugs cefepime (160μg/ml), ceft azidime (130 μg/ml), tobramycin (8 μg/ml), amikacin
(21 μg/ml) and ciprofloxacin (4,6 μg/ml) alone and amikacin+cefepime, amikacin+ceft azidime, amikacin+tobramycin,
ciprofloxacin+cefepime, ciprofloxacin+ceft azidime and ciprofloxacin+tobramycin combinations at therapeutic doses on polymorphonuclear
leucocyte (PMN) functions (phagocytosis and intracellular killing activity) of healthy volunteers whose average
age was 25, were investigated in vitro.
PMN’s 1x107cell/ml) were isolated by ficoll-hypaque gradient centrifugation method from venous blood with EDTA. Phagocytosis
and intracellular killing activity were assayed by modifying Alexander’s method.
Cefepime and amikacin did not change the PMN’s phagocytic and intracelular killing activities. Ceft azidime, tobramycin, ciprofloxacin significantly increased the PMN’s phagocytic activity (p<0,05, p<0,05, p<0,001) Th e increase in intracelular killing
activity was not statistically significant.
When antipseudomonal drugs combinations were compared with amikacin and ciprofloxacin alone, amikacin+ceft azidime
combination increased PMN’s intracelular killing activity, amikacin+tobramicin combination increased phagocytic activity
significantly (p<0,05). Amikacin + cefepime did not aff ect the PMN functions significantly. Ciprofloxacin+cefepime,
ciprofloxacin+ceft azidime, ciprofloxacin+tobramycin increased PMN’s phagocytic activity insignificantly (p>0,05).
