Microdilution Method Using “High Resolution ” Medium for Determining the Antifungal Susceptibility

This study was undertaken to evaluate the usefullness of high resolution medium in microdilution method when determining the susceptibilities of Candida albicans strains against antifungal agents. Hundred and five C.albicans strains were included in the study. The susceptibilities of the strains against amphotericin B and fluconazole were determined by microdilution method using two different media, RPMI 1640 medium buffered with MOPS according to NCCLS M27-A standards and high resolution medium buffered with phosphate buffer. The distribution of MIC values determined with two different media were compared with t test. At the same time overall results of both methods were considered to be in agreement when the MIC values were within +2 fold dilutions. On the other side the susceptibility catergories of the strains and the agreement rates were determined. No statistical difference was observed between the distribution of MIC values of two different media for amphotericin B and fluconazole (p=0.183 for amphotericin B and p=0.130 for fluconazole). The agreement rates of MIC values wtihin +2 fold dilutions were 82% and 89% for amphotericin B and fluconazole, respectively. These rates were detected to be 94 and 95% for amphotericin B and fluconazole when susceptibility categories were considered. According to our results, we conclude that high resolution media can be used in microdilution method for determining the susceptibilities of C.albicans strains against amphotericin B and fluconazole.
