Determination of autoantibodies by the indirect immunofluorescent (IIF) techniqe is the most preferred method for the diagnosis of connective tissue disorders. Differentiation of autoantibodies is the basis of correct diagnosis in otoimmun diseases. This method allows the extracted antigens (ENA) to be used for the differentiation of autoantibodies clearly.

In our study, we evaluated the results of Immunoblotting (IB) test in patients with positive anti-nuclear-antibody (ANA) which was determined by the IIF technique during one year period retrospectively. A total of 2587 patients were evaluated for ANA positivity by the IIF technique. Of the 2587 patients, 215 were found to be positive for +2< ANA by the IIF method, and they were studied by the IB test. Of the 215 patients with ANA positivity, 171 (79.5%) were found to have at least one autoantibody, whereas 44 (20.4%) were found to have no autoantibody by the IB test. The autoantibody against SS-A/Ro-52 antigen was the most frequent autoantibody determined by the IB test.
